Abstract
Inflammation is an evolutionally-selected protective mechanism. However, uncontrolled inflammation has deleterious consequences, characterized by tissue damage and disease onset. However, the genesis and regulation of each of disease-associated inflammation remain elusive. Macrophages, a special subset of immune cells, play a key role in orchestrating the initiation, progression and resolution of inflammation. However, tissue macrophages are highly heterogeneous and functionally plastic. The identification of new types of macrophages in pancreatic islets and their roles in controlling autoimmunity, metabolic process and beta cell function will be discussed. The goal is to develop new therapeutics for diabetes by modulating and harnessing macrophages.
